- 科研綜述
在牛胰島素人工合成研究中用鈉-氨還原胰島素硫硫鍵、除去苄基和其他保護基,並把處理後的A-、B-鏈還原並重氧化生產具有生物活力的胰島素提出併成功地實現了用天然肽於蛋白質結構功能研究在蛇肌果糖-1,6-二磷酸酯酶的研究中,測定了該酶的一級結構和晶體結構,克隆和表達了這個酶,系統地研究了該酶的動力學,作用機制和調節作用,發現該酶在反應中存在磷酰化中間物、別構部位和催化部位信息傳遞的分子基礎,發現ANP對蛇肌酶有激活和抑制雙重調節作用,提出了該酶催化過程的一個新的機制以及該酶在天然狀態下活性部位是不完善的觀點。在對果糖-6-磷酸-2-激酶/果糖-2,6一二磷酸酯酶的研究中,確定該酶的催化作用是雙底物雙產物序列催化機制,鎂離子有重要的調節作用,精氨酸殘基是底物結合的必需基團,以及該酶有一個非必需的活化基團確定了兔肝果糖二磷酸酯酶的催化和別構部位。此外,還證明了胸腺素β-4是由巨噬細胞而不是由胸腺細胞合成的。近年來,對於蛋白質的摺疊與去摺疊,結構域在摺疊和去摺疊中的作用,結構域在摺疊和去摺疊過程中的作用等方面進行了一系列的研究,並得到了一些重要的結果。

在半個世紀的科學生涯中,先生在人工全合成牛胰島素研究、蛋白質功能基因的修飾與其生物活性之間的定量關係研究、蛇肌果糖二磷酸酯酶研究、兔肝果糖-1,6-二磷酸酯酶研究、蛋白質的結構層次和蛋白質的摺疊研究等方面均取得了傲人成就,為研究所的發展和推動生化與分子生物學的國內交流及提升國內生化與分子生物學在國際上的影響力作出了重要的貢獻。 (上海生命科學研究院生物化學與細胞生物學研究所李林所長評)
