王娟學術成果

於2011年9月~2015年6月在愛爾蘭都柏林大學食品安全中心重點實驗室攻讀博士學位,主要從事細菌耐藥方面的研究,博士論文題目為《產超廣譜β內酰胺酶類大腸埃希菌內耐藥性質粒DNA基因序列的研究》,積累了細菌耐藥機理和傳播機制,分子生物學和信息生物學等研究方面的經驗,並取得了一定的成果。其中通過對16條分離自瑞士健康人羣和農場食源性動物中的產超廣譜β內酰胺酶(ESBLs)類大腸桿菌的耐藥質粒DNA全基因序列的比對研究發現,來自健康人體和農場食源性動物的耐藥性質粒具有相似的基因序列,所攜帶的抗生素耐藥基因擁有相似的基因環境。另外通過分子生物和信息生物學的方法發現,質粒不相容羣分型為IncI1,IncN,IncF和IncB/O的質粒類別,可以介導產超廣譜β內酰胺酶類基因 blaCTX-M-1和 blaTEM-52在健康人羣和農場食源性動物之間的傳播。基因插入序列IS Ecp1 和IS 26 ,作為重要的遺傳元件,可以增強 blaCTX-M-1基因的表達和介導其在不同質粒間的克隆傳播和水平傳播。另外,研究還首次報道了來自健康人體和食源性動物的耐藥質粒攜帶有相近的毒素-抗毒素系統。博士期間的研究結果首次報道了分離自瑞士健康人體和農場動物的大腸桿菌的質粒DNA序列,對瑞士人畜共患病和臨牀常用抗生素的耐藥基因的傳播途徑提供了有力證據,對減少畜牧業抗生素的使用提供了有力支持。
於2014年9月~2015年9月在都柏林大學食品安全中心重點實驗室做了為期一年的博士後研究,在博士後研究期間,以重要參與人身份參與了多項科研項目,其中包括愛爾蘭地區豬源性耐甲氧西林金黃色葡萄球菌(MRSA)的遺傳特性和耐藥特徵分析,嬰兒奶粉中阪崎腸桿菌的檢測和分析,奶源性腸球菌的基因分型和檢測等。在參與的愛爾蘭地區豬源性MRSA的遺傳特性和耐藥特徵分析的研究項目中,通過對愛爾蘭地區來自不同養殖場和不同成熟段豬體內分離的MRSA的基因分型和耐藥性的檢測,表明愛爾蘭地區MRSA具有多重耐藥的特點,研究結果為加強愛爾蘭地區生豬養殖重點地區豬源MRSA的檢測和流行傳播機制的研究奠定了基礎。
截止目前已發表中英文文章25篇,其中SCI 23篇,累計影響因子超過70分,其中第一作者署名文章共10篇(累計影響因子34.78分)。 [1]
