沈前華研究領域

植物如何識別病原菌並激活抗病,病原菌如何巧妙地躲避識別或抑制寄主抗性,專性寄生真菌如何與植物建立寄生關係等等是重要的生物學前沿問題。作物抗病性也是重要的穩產性狀,提高作物品種的抗病性可以減少農藥使用、保護環境、綠色生產。實驗室採用重要糧食作物大小麥與白粉菌分子互作的研究體系,採用遺傳學、細胞生物學、生物化學和組學等研究手段研究作物抗病的分子機制,研究重要病原真菌的致病分子機理, 挖掘新型抗病基因,為抗病育種提供理論基礎,並應用於創制抗病育種材料。
1、植物免疫受體的抗病機制
研究抗病蛋白的抗病機制,探索抗病基因在抗病育種中的應用。抗病蛋白或NLR免疫受體介導對病原菌專化性識別並誘導抗病免疫反應,我們研究這類抗病蛋白誘導抗病及細胞死亡的分子機制,以及抗病激活與轉錄重編程的關係,鑑定抗病蛋白複合體組份及下游信號組份,闡明抗病基因轉錄及翻譯後的調控機制。
2、植物與病原真菌互作的跨界信號
病原真菌(如白粉菌)編碼數百個效應蛋白,大多被分泌到寄主細胞中,通過干擾寄主的靶標蛋白功能抑制植物的抗病免疫反應增強病原菌致病性。植物和病原真菌的小分子RNA(主要是miRNA、siRNA)在植物與病原菌互作中也發揮重要的調控作用,並通過跨界調控靶標基因的表達起調控作用。通過研究效應蛋白的功能和作用機制,鑑定sRNA以及靶標基因及其調控機制,闡明跨界sRNA的作用機制。
3、麥類作物抗病遺傳及抗病基因克隆
麥類作物真菌病害主要包括白粉病、鏽病和赤黴病。我們研究大、小麥對病原真菌的抗病遺傳,篩選抗病種質資源、挖掘抗病新位點,通過GWAS等技術鑑定和克隆新型抗病基因,包括廣譜和專化性抗病基因。
