在上一集故事中，尋寶專家本·蓋茨終於解開了《獨立宣言》的寶藏之謎，不但贏 得了美人的芳心，還得到了國家饋贈的鉅額獎金……

如今，志得意滿的本也遇到了那麼點小麻煩，由於他致力於全世界地遊蕩，就為了挖掘出被隱藏起來的歷史和寶藏，然而，本的女友阿比蓋爾·蔡斯卻以保護一切與歷史有關的遺蹟為己任，兩人之間的分歧越來越大，已經演變成了無休止的爭吵之後的階段性冷戰，本甚至收拾行囊，離開兩人的愛巢，搬到父親帕特里克那裏去暫住了。

亞伯拉罕·林肯的死一直以來都是美國曆史上最慘重的損失，雖然兇手約翰·沃克斯·布斯隨即被揪了出來，可是在他住所搜到的日記卻無端地少了18頁，為本來已經明瞭的案情平添一層神秘的面紗。最讓人百思不得其解的是，100多年後今天，18頁失蹤的日記其中的一頁卻突然現身，學者傑布·威爾金森對其中一些曖昧不明的説詞解釋為暗殺信息，不幸的是，這頁日記中所有的一切，似乎都在指明一個事實，那就是本的曾曾祖父很可能是林肯刺殺一案背後真正的策劃與主謀，蓋茨家顯赫的聲譽蒙上了一層陰影。

為了證明自己的祖先當時只是在解碼一個藏寶圖，而非謀殺總統，本找來正與國税局 糾纏不清的老搭檔賴利·普爾，聯同父親帕特里克和互相看不順眼的女友阿比蓋爾，一起去尋找傳説中的那座“金城”，用事實來還給他的曾曾祖父一個清白。然而線索太多也太分散，將他們從巴黎帶到倫敦，甚至還潛入位於白金漢宮的女王私人起居室。不過，他們最後重新又回來了美國，那個一切開始的地方，本甚至不惜綁架現在的總統，只為了去看看只有總統本人才能翻看、記錄的“秘密之書”……最糟糕的是，本還驚動了他的母親艾米麗，一位著名的古英語學者，而艾米麗在32年前與帕特里克就尋寶的側重點發生了激烈的爭吵之後，就再沒有説過一句話。這場同樣充滿了驚險刺激的旅程，不但幫助本和他的同伴們追蹤到了他們想知道的事實真相，還發現了這個世界上最珍貴的秘密遺蹟。

繼發現了聖殿騎士團的財富後，已成為世界最有名的寶藏獵人本·蓋茨(尼古拉斯·凱奇飾)與身為大學教授的父親帕特里克·蓋茨(喬·沃特飾)又發現了暗殺林肯的刺客約翰·威爾克斯·布斯的一頁日記。布斯被打死後在他身上找到了他的日記，但有幾頁被撕掉了，一直沒有找到。這次本發現的就是其中的一頁。讓他們大為驚訝的是，這頁日記的內容居然暗示他們的祖先托馬斯·蓋茨是林肯總統刺殺案的同謀。

本攜手身為國家檔案保管員的前女友艾比蓋爾·查斯(戴安·克魯格飾)，和老搭檔——電腦高手萊利·普爾(賈斯汀·巴薩飾)踏上奔波世界各地的歷險旅程，其間險象環生。來自華盛頓特區、巴黎、倫敦和美國中心地帶等多處秘密中心的危機交疊在一起。

本、帕特里克、艾比蓋爾和賴利小心翼翼地追蹤線索，這有可能徹底翻轉歷史以及他們的生活。從尋求歷史真相擴展到尋找一本被嚴加保護的堪稱世上最神秘的書，並由此找到一筆聞名的神秘財富。帕特里克萬萬沒想到本居然搬出了一個秘密武器——就是他令人敬畏的母親，帕特里克的前妻，語音學家艾米麗·阿普爾頓(海倫·米倫飾)。她和帕特里克已經32年沒説過話了。她很快身陷其中，不過他們幾個人並不是唯一尋找真相的小團隊。那個讓本注意到這一頁日記的人叫米奇·威爾金森(艾德·哈里斯飾)。他自己也有不為人知的家族秘密。他的野心並不僅限於尋找寶藏，別有用心的他與本一干人馬的生死衝突跌宕起伏。

堅毅號辦公桌的故事是真的，當然除了密碼那一部分，影片中桌子裏的機關全是電腦製作

·因為在《女王》中的完美演繹，海倫·米倫得到許可會見伊麗莎白二世女王，不過由於她正在南達科塔拍攝這部影片，錯過了這個機會。

·影片開拍了幾個月後，攝影師阿米爾·M·莫卡利(Amir M.　Mokri)的位置被約翰·施瓦茲曼(John Schwartzman)取代。
